electrospray ionisation comes up often in conversation and rarely with the context attached. Here we lay out the basics in order, then work through the practical considerations.

Compared with older secretagogues such as hexarelin or GHRP-6, ipamorelin shows weaker stimulation of cortisol, prolactin, and appetite in the animal models used for early characterization. Whether that selectivity is preserved across longer human exposures remains an open question, because published clinical data are limited in size and duration. Reported effects on food intake are generally described as modest. The compound is therefore treated in the literature as a relatively selective research tool rather than a fully characterized therapeutic agent.
Ipamorelin is a synthetic pentapeptide that belongs to the growth hormone secretagogue class of compounds. Researchers at a pharmaceutical company first described it in the 1990s while screening small peptides for growth hormone releasing activity. Its chain contains five amino acid residues, two of which are non-natural building blocks, including 2-aminoisobutyric acid and a naphthylalanine derivative. The molecule was designed to act at the ghrelin receptor while avoiding several effects observed with earlier secretagogues.
At the cellular level, ipamorelin binds the growth hormone secretagogue receptor, also called the ghrelin receptor. Activation of this receptor on pituitary somatotroph cells triggers a signaling cascade that leads to release of growth hormone into circulation. Because release follows a pulsatile pattern, studies often report peak concentration and total area under the curve rather than a single time point. Selectivity for this receptor is the property most frequently discussed in comparative work.
The compound was developed at Novo Nordisk during the 1990s as part of a programme seeking secretagogues with improved selectivity. It was described in the peer-reviewed literature in 1998 alongside related pentapeptides from the same series. Investigators advanced it because it raised growth hormone output in animal models while leaving other pituitary hormones comparatively unaffected. The development code NNC 26-0161 appears in earlier reports, and ipamorelin later became the common designation in published work.
Selectivity is the property most often attached to this peptide. Published animal and early human studies record increases in growth hormone release after administration, with adrenocorticotropic hormone and cortisol responses remaining small by comparison. Effects on appetite-related pathways also appear weaker than those reported for several earlier secretagogues. Reviews that compare members of the growth hormone secretagogue family cite these findings frequently, though the receptor-level explanation for the selectivity continues to be debated rather than settled.

Identity and purity are assessed by complementary methods rather than a single test. Reversed-phase high-performance liquid chromatography separates the peptide from related impurities and reports a percentage purity. Mass spectrometry, most often with electrospray ionization, confirms the expected molecular mass and detects sequence-related variants. Amino acid analysis can verify composition, while water content and residual counterion measurements support the mass balance of a batch. Stability studies under accelerated conditions are used to estimate shelf life, though such estimates carry uncertainty for long-term storage.
Material supplied for research use is normally a white to off-white lyophilized powder. The solid is hygroscopic and is handled in a low-humidity environment to limit water uptake. Bulk quantities are frequently shipped in sealed vials under inert gas. Once reconstituted in water or a neutral buffer, the solution is less stable than the dry powder and is usually divided into single-use aliquots.
The lyophilised solid is normally held at -20 °C or colder, shielded from light and moisture. Stability in that state is measured in years, although shelf life depends on residual water content and the container seal. Once dissolved, the peptide is more fragile: aqueous solutions are commonly kept at 2-8 °C and used within days to weeks, and repeated freeze-thaw cycling is avoided. Strongly acidic or basic conditions accelerate hydrolysis, and prolonged exposure to them can strip the terminal amide.
Purity assessment for this peptide relies mainly on reversed-phase high-performance liquid chromatography. A C18 column with a water-acetonitrile gradient containing trifluoroacetic acid separates the target from truncated sequences and oxidation products. Detection near 214 nm exploits the amide backbone, while the aromatic side chains allow additional monitoring close to 280 nm. Reported purity values depend on the method, so a certificate of analysis carries weight only when gradient, column and integration parameters are given.
== Legacy == Since his retirement in 2003, Austin has been widely regarded and cited as one of the greatest and most influential professional wrestlers of all time. Sports Illustrated ranked him third on their top 101 greatest wrestlers of all-time list. In 2020, SPORTbible ranked Austin as the greatest wrestler of all time, and the following year in 2021 Bleacher Report also named him the greatest WWE wrestler of all time. He has been described as the most influential wrestler in Raw history, and the poster boy for the Attitude Era. Several former world champions have named Austin as part of their "Mount Rushmore" of wrestling, including The Rock, The Undertaker, Hulk Hogan, Ric Flair, and John Cena, and a 2012 poll conducted by WWE saw Austin picked second on a fan voted version of the concept. When Vince McMahon inducted Austin into the WWE Hall of Fame in 2009, he referred to Austin as "the greatest WWE superstar of all time". Austin was the biggest box office draw in WWE since Hulk Hogan. His contributions in saving the WWF and winning the Monday Night Wars against WCW helped future superstars like John Cena, who would go onto establish WWE as a global brand. He headlined WrestleMania X-Seven, the first WrestleMania to achieve over 1 million buys. The event was universally acclaimed and is often regarded as the greatest pay-per-view in professional wrestling history. It is also noted as the pinnacle of the Attitude Era, occurring just a week after the WWF bought out their competition, WCW. During his early years as a wrestler, Austin was a technical wrestler.
==== The locus of power and organized crime ==== Some espouse that all organized crime operates at an international level, though there is currently no international court capable of trying offenses resulting from such activities (the International Criminal Court's remit extends only to dealing with people accused of offenses against humanity, e.g., genocide). If a network operates primarily from one jurisdiction and carries out its illicit operations there and in some other jurisdictions it is 'international,' though it may be appropriate to use the term 'transnational' only to label the activities of a major crime group that is centered in no one jurisdiction but operating in many. The understanding of organized crime has therefore progressed to combined internationalization and an understanding of social conflict into one of power, control, efficiency risk and utility, all within the context of organizational theory. The accumulation of social, economic and political power have sustained themselves as a core concerns of all criminal organizations:
=== Lung transplantation === Lung transplantation may be suitable for those patients physically eligible to undergo a major transplant operation. In IPF patients, lung transplant has been shown to reduce the risk of death by 75% as compared with patients who remain on the waiting list. Since the introduction of the lung allocation score (LAS), which prioritizes transplant candidates based on survival probability, IPF has become the most common indication for lung transplantation in the USA. Symptomatic patients with IPF younger than 65 years of age and with a body mass index (BMI) ≤26 kg/m2 should be referred for lung transplantation, but there are no clear data to guide the precise timing for LTx. Although controversial, the most recent data suggest that bilateral lung transplantation is superior to single lung transplantation in patients with IPF. Five-year survival rates after lung transplantation in IPF are estimated at between 50 and 56%.
Pellagra was first reported in 1902 in the United States, and has "caused more deaths than any other nutrition-related disease in American history", reaching epidemic proportions in the American South during the early 1900s. Poverty and consumption of corn were the most frequently observed risk factors, but the exact cause was not known, until groundbreaking work by Joseph Goldberger. A 2017 National Bureau of Economic Research paper explored the role of cotton production in the emergence of disease; one prominent theory is that "widespread cotton production had displaced local production of niacin-rich foods and driven poor Southern farmers and mill workers to consume milled Midwestern corn, which was relatively cheap but also devoid of the niacin necessary to prevent pellagra." The study provided evidence in favor of the theory: there were lower pellagra rates in areas where farmers had been forced to abandon cotton production (a highly profitable crop) in favor of food crops (less profitable crops) due to boll weevil infestation of cotton crops (which occurred randomly). Pellagra developed especially among the vulnerable populations in institutions such as orphanages and prisons, because of the monotonous and restricted diet. Soon pellagra began to occur in epidemic proportions in states south of the Potomac and Ohio rivers. The pellagra epidemic lasted for nearly four decades beginning in 1906. It was estimated that there were 3 million cases, and 100,000 deaths due to pellagra during the epidemic.

== Composition == Most hotspot volcanoes are basaltic (e.g., Hawaii, Tahiti). As a result, they are less explosive than subduction zone volcanoes, in which water is trapped under the overriding plate. Where hotspots occur in continental regions, basaltic magma rises through the continental crust, which melts to form rhyolites which can form violent eruptions. For example, the Yellowstone Caldera was formed by some of the most powerful volcanic explosions in geologic history. However, when the rhyolite is completely ejected, it may be followed by eruptions of basaltic magma rising through the same lithospheric fissures (cracks in the lithosphere). An example of this activity is the Ilgachuz Range in British Columbia, which was created by an early complex series of trachyte and rhyolite eruptions, and late extrusion of a sequence of basaltic lava flows. The hotspot hypothesis is now closely linked to the mantle plume hypothesis. The detailed compositional studies now possible on hotspot basalts have allowed linkage of samples over the wider areas often implicate in the later hypothesis, and its seismic imaging developments.
== History == "Enthesis" is rooted in the Ancient Greek word, "ἔνθεσις" or "énthesis," meaning "putting in," or "insertion." This refers to the role of the enthesis as the site of attachment of bones with tendons or ligaments. Relatedly, in muscle terminology, the insertion is the site of attachment at the end with predominant movement or action (opposite of the origin). Thus the words (enthesis and insertion [of muscle]) are proximal in the semantic field, but insertion in reference to muscle can refer to any relevant aspect of the site (i.e., the attachment per se, the bone, the tendon, or the entire area), whereas enthesis refers to the attachment per se and to ligamentous attachments as well as tendinous ones.
Context-sensitive half-life or context sensitive half-time is defined as the time taken for blood plasma concentration of a drug to decline by one half after an infusion designed to maintain a steady state (i.e. a constant plasma concentration) has been stopped. The "context" is the duration of infusion. When a drug which has a multicompartmental pharmacokinetic model is given by intravenous infusion it initially will distribute to the central compartment and then move out of this compartment into one or two peripheral compartments. Once this infusion is discontinued, drug continues to move into the peripheral compartments until an equilibrium is reached. At this time, the only way drug may leave plasma is by metabolism or excretion. As the plasma concentration falls, the concentration gradient of drug reverses and drug moves from peripheral compartments back into plasma, maintaining the plasma concentration of the drug, often prolonging the pharmacological effect. If an infusion has reached steady state then the context-sensitive half-life is equal to the terminal plasma half-life of the drug. Otherwise it will be shorter than the terminal elimination half-life. Remifentanil is relatively context insensitive whilst fentanyl and thiopentone are examples of drugs which have significant context-sensitive changes in their half-life. The Context-Sensitive Half-Time describes the time required for the plasma drug concentration to decline by 50% after terminating an infusion of a particular duration.
Some effects are dependent upon the route of administration, with intravenous use associated with the transmission of many disease such as hepatitis C, HIV/AIDS, and potential medical emergencies such as infection, thrombosis, or pseudoaneurysm, while inhalation may be associated with increased lower respiratory tract infection, lung cancer, and pathological restricting of lung tissue. Cocaine may also increase risk for autoimmune disease and damage nasal cartilage. Abuse of methamphetamine produces similar effects as well as marked degeneration of dopaminergic neurons, resulting in an increased risk for Parkinson's disease.

== Medical uses == The two most commonly used methods of confirming allergen sensitization are skin testing and allergy blood testing. Both methods are recommended by the NIH guidelines and have similar diagnostic value in terms of sensitivity and specificity. Advantages of the allergy blood test range from: excellent reproducibility across the full measuring range of the calibration curve, it has very high specificity as it binds to allergen specific IgE, and extremely sensitive too, when compared with skin prick testing. In general, this method of blood testing (in-vitro, out of body) vs skin-prick testing (in-vivo, in body) has a major advantage: it is not always necessary to remove the patient from an antihistamine medication regimen, and if the skin conditions (such as eczema) are so widespread that allergy skin testing cannot be done. Allergy blood tests, such as ImmunoCAP, are performed without procedure variations, and the results are of excellent standardization. Adults and children of any age can take an allergy blood test. For babies and very young children, a single needle stick for allergy blood testing is often more gentle than several skin tests. However, skin testing techniques have improved. Most skin testing does not involve needles and typically skin testing results in minimal patient discomfort. Drawbacks to RAST and ImmunoCAP techniques do exist. Compared to skin testing, ImmunoCAP and other RAST techniques take longer to perform and are less cost effective.
In 2016, a set of 355 genes likely present in the LUCA was identified. A total of 6.1 million prokaryotic genes from Bacteria and Archaea were sequenced, identifying 355 protein clusters from among 286,514 protein clusters that were probably common to the LUCA. The results suggest that the LUCA was anaerobic with a Wood–Ljungdahl (reductive Acetyl-CoA) pathway, nitrogen- and carbon-fixing, thermophilic. Its cofactors suggest dependence upon an environment rich in hydrogen, carbon dioxide, iron, and transition metals. Its genetic material was probably DNA, requiring the 4-nucleotide genetic code, messenger RNA, transfer RNA, and ribosomes to translate the code into proteins such as enzymes. LUCA likely inhabited an anaerobic hydrothermal vent setting in a geochemically active environment. It was evidently already a complex organism, and must have had precursors; it was not the first living thing. The physiology of LUCA has been in dispute. Previous research identified 60 proteins common to all life. Metabolic reactions inferred in LUCA are the incomplete reverse Krebs cycle, gluconeogenesis, the pentose phosphate pathway, glycolysis, reductive amination, and transamination.
Olanzapine/fluoxetine (trade name Symbyax, created by Eli Lilly and Company) is a fixed-dose combination medication containing olanzapine (Zyprexa), an atypical antipsychotic, and fluoxetine (Prozac), a selective serotonin reuptake inhibitor (SSRI). Olanzapine/fluoxetine is primarily used to treat the depressive episodes of bipolar I disorder as well as treatment-resistant depression.
=== Mechanism of action === The alkylation mechanism of melphalan is shared by all nitrogen mustards, proceeding through the formation of an aziridinium cation. This highly reactive intermediate interacts with nitrogen atoms in nucleotide bases, generating positively charged adducts. Melphalan chemically modifies DNA nucleotides through alkylation, primarily targeting guanine at the N7 position within the major groove, and to a lesser extent adenine at the N3 position within the minor groove.

It is a synthetic pentapeptide in the growth hormone secretagogue family. The chain contains five residues, two of which are non-natural amino acids.
It acts as an agonist at the growth hormone secretagogue receptor, also known as the ghrelin receptor. Binding at pituitary somatotroph cells promotes growth hormone release.
Early studies report weaker effects on cortisol, prolactin, and appetite than compounds such as GHRP-6. The size of that difference in humans is not firmly established.
It is a synthetic five-amino-acid peptide that acts as a growth hormone secretagogue. Three of its residues are non-standard amino acids, and the chain ends in an amide rather than a free acid. The molecule is small enough that it can be characterised by routine peptide analytical techniques.
